Description

A mouse cell isolation kit from STEMCELL Technologies uses immunomagnetic positive selection to isolate CD138+ cells from single-cell suspensions of mouse splenocytes or bone marrow. The column-free workflow relies on an EasySep™ magnet, with unwanted cells removed by pouring off the supernatant while target cells remain in the tube. The kit is supplied for processing 2 x 10^9 cells.

A key advantage is the ability to release bound magnetic particles (EasySep™ Releasable RapidSpheres™) following positive selection, yielding highly purified, particle-free cells immediately ready for downstream applications such as flow cytometry, cell culture, or hybridoma generation. The kit achieves up to 95% purity of plasma cells from immunized mouse samples and is compatible with hybridoma generation protocols, including electrofusion. Reported plasma cell (CD138+CD267 (TACI)+) purity in the isolated fraction is 77.2 ± 2.4% from naïve C57BL/6 mouse splenocytes, 86.1 ± 7.4% from immunized BALB/c mouse splenocytes, 62.0 ± 13.1% from naïve C57BL/6 mouse bone marrow, and 85.5 ± 9.8% from immunized BALB/c mouse bone marrow (mean ± SD, using the purple EasySep™ Magnet).

Kit components include EasySep™ Release Mouse CD138 Positive Selection Cocktail, EasySep™ Mouse FcR Blocker, EasySep™ Isolation Cocktail Enhancer, EasySep™ Releasable RapidSpheres™, and EasySep™ Release Buffer (Concentrate). All components are stored at 2 - 8°C and should not be frozen. Following isolation, antibody complexes remain bound to the cell surface and may interact with Brilliant Violet™ antibody conjugates, polyethylene glycol-modified proteins, or other chemically related ligands. When using single-cell suspensions from other tissue types, this kit may require optimization.
