EasySep Release Ms Biotin Pos Slctn Kit

From STEMCELL Technologies, this kit isolates highly purified, magnetic particle-free cells labeled with biotinylated antibodies from mouse splenocytes in less than 40 minutes. Cells labeled with biotinylated antibodies (not provided) bind to antibody complexes recognizing biotin and to EasySep Releasable RapidSpheres, then are separated without columns using an EasySep magnet. Unwanted cells are poured off while desired cells remain in the tube. A release buffer step then removes the magnetic particles, leaving antibody complexes on the cell surface and the cells immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction.

The kit is supplied with a biotin positive selection cocktail, Releasable RapidSpheres, release buffer concentrate, and an EasySep Mouse FcR Blocker, which is supplied only with this mouse version of the kit. All components should be stored at 2 - 8°C and not frozen; they are stable until the expiry date on the label. Recommended medium is EasySep Buffer or PBS containing 2% FBS and 1 mM EDTA, free of Ca++ and Mg++. For mouse spleen preparation, the spleen is disrupted in recommended medium, the suspension is passed through a 70 μm mesh nylon strainer, centrifuged at 300 x g for 10 minutes, and resuspended at 1 x 10^8 nucleated cells/mL.

Typical Cell Isolation Profile from splenocytes: starting with mouse splenocytes, the purities of the start and final isolated fractions are 58.4% and 95.1%, respectively, using a biotinylated anti-mouse CD19 antibody, as assessed by labeling with CD19 and CD45R/B220. The kit is designed for processing 1 x 10^9 cells.
