EasySep Release Mouse PE Pos Slctn Kit

STEMCELL Technologies offers an immunomagnetic kit designed to isolate highly purified mouse cells labeled with PE (phycoerythrin)-conjugated antibodies, typically from mouse splenocytes. The kit is intended for processing 1 x 10^9 cells and employs particle release technology, producing magnetic particle-free, antibody-bound target cells in less than 40 minutes.

Cells are labeled with user-supplied PE-conjugated antibodies, then incubated with a positive selection cocktail and Releasable RapidSpheres before separation using an EasySep magnet without columns. Unwanted cells are simply poured off while the labeled cells remain in the tube. A release buffer then detaches the magnetic particles from the isolated cells, which are immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction. Antibody complexes may remain bound to the cell surface and could interact with Brilliant Violet antibody conjugates, polyethylene glycol-modified proteins, or other chemically related ligands.

Included components comprise the Release PE Positive Selection Cocktail, Releasable RapidSpheres, Release Buffer Concentrate, and Mouse FcR Blocker, all stored at 2 - 8°C. In a representative isolation from mouse splenocytes using a PE-conjugated anti-mouse CD19 antibody, start and final purities of 59.1% and 98.6% have been observed. In a mouse lung application using PE-conjugated anti-mouse CD326 (EpCAM) alongside biotin-conjugated CD45 antibodies, EpCAM+CD45- cells achieved 86% purity.
