EasySep Release Hu CD45 Pos Slctn Kit

From STEMCELL Technologies, this kit enables isolation of highly purified and magnetic particle-free human CD45+ leukocytes, including tumor-infiltrating leukocytes (TILs), from single-cell suspensions of primary human tissues and tumors. The positive selection approach uses EasySep™ Releasable RapidSpheres™ and antibodies recognizing the CD45 surface marker, with separation performed without columns using an EasySep™ magnet. Unwanted cells are poured off while desired cells remain in the tube, and bound magnetic particles are subsequently removed so the isolated CD45+ cells are immediately available for downstream applications such as flow cytometry, culture, and DNA/RNA extraction.

The procedure delivers highly purified human CD45+ leukocytes and TILs in less than 45 minutes, with a no-wash removal step for the magnetic particles. Following isolation with this EasySep™ Release kit, antibody complexes remain bound to the cell surface and may interact with Brilliant Violet™ antibody conjugates, polyethylene glycol-modified proteins, or other chemically related ligands. The kit is designed for processing 1 x 10^9 cells and is supplied with a Selection Cocktail (a combination of monoclonal antibodies in PBS with 0.1% BSA), Releasable RapidSpheres™ (a suspension of magnetic particles in water), and Release Buffer Concentrate. Components should be stored at 2 - 8°C, must not be frozen, and are stable until the expiry date on the label.

The selection reagent is described as a mouse monoclonal IgG1 antibody against human and chimpanzee CD45, supporting column-free immunomagnetic positive separation of CD45+ cells.
