Immunomagnetic positive selection combined with post-selection particle release enables researchers to recover highly purified, particle-free human CD19+ B lymphocytes from heterogeneous samples. This release format from STEMCELL Technologies is engineered for downstream applications where residual antibody-particle complexes on the cell surface could otherwise interfere with functional or staining readouts.

The kit targets the pan-B cell marker CD19 with a human-specific antibody cocktail and is compatible with whole blood, leukapheresis products, bone marrow, and peripheral blood mononuclear cell preparations. Isolated CD19+ cells maintain viability and surface marker integrity, supporting downstream flow cytometry, cell culture, in vitro functional assays, and adoptive transfer studies.

Each kit contains a Selection Cocktail, Release Buffer, and Magnetic Particles supplied in individual vials. Components ship at ambient temperature on cold packs and should be refrigerated upon arrival; storage at 4°C preserves reagent activity through the printed expiration date.

For processing, a 50-200 uL sample volume of whole blood (or proportional volumes of buffy coat or mononuclear cell preparations) is incubated with the antibody cocktail, followed by addition of magnetic particles. After a short incubation, the tube is placed in an EasySep magnet for column-free separation. The Release Buffer is then applied to detach magnetic particles from the selected CD19+ population, yielding highly purified, particle-free cells. The entire workflow is performed in standard 5 mL polystyrene tubes without columns.

Released CD19+ B cells obtained with this kit are validated for downstream protocols including flow cytometry staining, proliferation assays, immunoglobulin production studies, and B cell receptor repertoire analysis. All kit components are for research use only and are not intended for diagnostic, therapeutic, or clinical applications in patients.
