EasySep Rat B Cell Isolation Kit

A column-free immunomagnetic negative selection kit from STEMCELL Technologies designed to isolate untouched, highly purified B cells from single-cell suspensions of rat splenocytes, lymph nodes, or whole blood. The kit targets non-B cells for removal with antibodies recognizing specific cell surface markers; unwanted cells are labeled with antibodies and magnetic particles, then separated using an EasySep™ magnet, leaving the desired cells to be poured off into a new tube. Isolated cells are immediately available for downstream applications such as flow cytometry, cell culture, or DNA/RNA extraction.

The kit is rated for processing up to 1 x 10^9 cells. When starting with a single-cell suspension of rat splenocytes, the B cell content (CD45RA+CD3-) of the isolated fraction is typically 94.1 ± 3.8% (mean ± SD using the purple EasySep™ Magnet), and the product literature reports up to 95% purity. In one example, the start and final isolated fractions measured 44.5% and 95.6% purity, respectively. The kit has been verified for use with the Sprague Dawley strain and is expected to also be compatible with other strains. Purity assessment by flow cytometry is recommended using anti-rat CD45RA and anti-rat CD3 antibodies.

Components include the EasySep™ Rat B Cell Isolation Cocktail (a combination of monoclonal antibodies in PBS) and EasySep™ Dextran RapidSpheres™ (a suspension of magnetic particles in water), both stored at 2 - 8°C. The protocol is fast and column-free, and ammonium chloride treatment is not recommended when preparing the cells for separation.
