Overview

Magnetic positive selection of specific cell populations from mouse tissue or blood samples is the core application of this EasySep kit from STEMCELL Technologies. The workflow begins with a user-supplied PE-conjugated antibody directed at the target antigen, followed by the kit's selection cocktail and dextran-coated magnetic nanoparticles. After a brief room-temperature incubation in a single tube, the sample is placed inside an EasySep magnet, where unlabeled cells are poured off and the desired PE-labeled fraction is retained on the tube wall.

This second-generation Kit II format completes most separations in approximately 5 minutes without requiring separation columns, minimizing cell stress and handling steps. Compatible starting materials include mouse spleen, bone marrow, lymph node, thymus, and peripheral blood, typically processed at 0.25 to 2 mL working volumes per sample. Recovered cells remain in suspension and are immediately ready for downstream applications such as flow cytometry, in vitro culture, RNA sequencing, and adoptive transfer studies in vivo.

Because end users select their own PE-conjugated detection reagent, the kit is flexible and can be paired with any of the many commercially available mouse-specific PE antibodies. Isolated cells are free of magnetic particles after the procedure, making them suitable for functional assays and transplantation into recipient mouse models.

Intended for research use only. Supplied as a single kit containing single-use reagents; store refrigerated to maintain reagent performance across the indicated shelf life.
