EasySep Ms MDSC Neg Iso Kit

This immunomagnetic negative selection kit from STEMCELL Technologies isolates untouched and highly purified CD11b+Gr1+ myeloid-derived suppressor cells (MDSCs) from mouse splenocytes, bone marrow, or peripheral blood in as little as 18 minutes. The kit targets non-MDSCs for removal using biotinylated antibodies and streptavidin-coated magnetic particles, then separates unwanted cells without columns using an EasySep™ magnet. Desired cells are simply poured off into a new tube and remain immediately available for downstream applications.

Up to 96% purity can be achieved. Starting with 4T1 tumor-bearing BALB/c mouse splenocytes, the MDSC content (CD11b+Gr1+) of the isolated fraction is typically 94.3 ± 2.1% (mean ± SD) using the purple EasySep™ Magnet. Starting with naïve C57BL/6 mouse splenocytes, the CD11b+Gr1+ cell content of the isolated fraction is typically 86 ± 4.6% (mean ± SD) using the same magnet. Isolated cells are suitable for flow cytometry, culture, or cell-based assays.

Three components are supplied: an Isolation Cocktail (a combination of monoclonal antibodies in PBS and 0.1% BSA), Streptavidin RapidSpheres™ (a suspension of magnetic particles in PBS), and a Mouse FcR Blocker. All components should be stored at 2 - 8°C and not frozen. The kit is designed for processing 1 x 10^9 cells.
