EasySep Mouse Plasmacytoid DC Iso Kit

From STEMCELL Technologies, this immunomagnetic negative selection kit isolates untouched and highly purified plasmacytoid dendritic cells (pDCs; PDCA-1+CD11c+) from mouse splenocytes. The procedure is fast, easy-to-use, and column-free, with up to 94% purity. Non-pDCs are targeted for removal using biotinylated antibodies recognizing specific cell surface markers, and unwanted cells are labeled and separated without columns using an EasySep™ magnet. Desired cells are simply poured off into a new tube and are immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction.

The kit is designed to process up to 2 x 10^9 cells and supports manual protocols using EasySep™ magnets as well as fully automated separation using the RoboSep™ instrument. Recommended medium includes EasySep™ Buffer, RoboSep™ Buffer, or PBS containing 2% FBS and 1 mM EDTA. When starting with mouse splenocytes, the pDC content (PDCA-1+CD11c+) of the enriched fraction typically ranges from 62 - 94%. Components include an Isolation Cocktail (a combination of monoclonal antibodies in PBS and 0.1% BSA, including an Fc receptor blocking antibody), a Biotin Selection Cocktail, and D2 Magnetic Particles, all stored at 2 - 8°C.
