EasySep Mouse Pan-Naïve T Cell Iso Kit

STEMCELL Technologies offers a column-free immunomagnetic negative selection kit for isolating untouched, highly purified pan-naïve T cells (CD3+CD44-/lowCD62Lhigh) from mouse splenocytes. The procedure uses biotinylated antibodies recognizing specific cell surface markers—targeting unwanted non-naïve T cells expressing CD11b, CD19, CD24, CD25, CD44, CD45R/B220, CD49b, and TER119—which are then labeled with streptavidin-coated magnetic particles and separated without columns using an EasySep™ magnet. Desired cells are simply poured off into a new tube and are immediately available for downstream applications such as flow cytometry, culture, or cell-based assays.

The kit is designed for processing 1 x 10^9 cells and achieves up to 97% purity, with the pan-naïve T cell content of the isolated fraction typically ranging from 90 - 97% when starting with splenocytes from an uninfected mouse. It is fast and easy-to-use, yielding untouched, viable cells. Single-cell suspensions from other tissue types may require optimization. The workflow is compatible with multiple magnet platforms from STEMCELL Technologies, including the EasySep™ magnet, The Big Easy, EasyPlate™, EasyEights™, and the fully automated RoboSep™ system.
