EasySep Mouse Pan-B Cell Isolation Kit

A column-free immunomagnetic kit from STEMCELL Technologies enables isolation of untouched and highly purified mouse pan-B cells (CD19+, CD19+CD138+, and CD138+), including conventional B-2 B cells, B-1 B cells, and plasma cells, from mouse splenocytes by negative selection. When using single-cell suspensions from other tissue types, the kit may require optimization.

The procedure targets non-pan-B cells for removal using biotinylated antibodies and streptavidin-coated magnetic particles; unwanted cells are retained in an EasySep™ magnet while the desired cells are poured off into a new tube and become immediately available for downstream applications such as flow cytometry, culture, or cell-based assays. Starting with C57BL/6 mouse splenocytes, the pan-B cell content of the isolated fraction typically ranges from 91 - 98%.

Both manual EasySep™ magnet protocols and a fully automated RoboSep™ workflow are supported. B cells isolated with the kit have also been combined with the ImmunoCult™ Mouse B Cell Expansion workflow, where they expanded 176.9 ± 29.8-fold after 9 days of culture.
