EasySep Mouse ILC2 Enrichment Kit - Sam

Murine group 2 innate lymphoid cells (ILC2s) drive type-2 cytokine responses at mucosal barriers, and their low abundance makes enrichment essential before flow cytometry or transcriptional analysis. This negative-selection kit from STEMCELL Technologies uses biotinylated antibodies plus magnetic particles to deplete non-ILC2 lineages, leaving target cells untouched, viable, and ready for downstream assays in roughly 30 minutes.

The workflow requires no columns: after a brief antibody incubation, samples are placed in an EasySep magnet for 3 minutes per round, and untouched ILC2s are simply pipetted into a fresh tube. Compatible buffers include PBS supplemented with 2% fetal bovine serum, and single-cell suspensions prepared from lung, spleen, mesenteric lymph node, or small intestinal lamina propria digests work in the protocol. The kit tolerates fresh and cryopreserved inputs at scales up to 1 x 10^9 cells per preparation, with no visible effect on surface-marker or transcription-factor profiles.

Intended for research use only, the reagents ship in a single-use format consisting of a 1 mL antibody cocktail vial and a 1 mL magnetic particle suspension. Purified ILC2s remain amenable to in vitro restimulation with PMA and ionomycin for intracellular cytokine staining, IL-5 and IL-13 secretion profiling, or adoptive transfer into mouse models of allergic airway inflammation and helminth infection.
