EasySep Mouse/Human Chimera Iso Kit

STEMCELL Technologies offers this immunomagnetic negative selection kit for isolating untouched, highly purified human cells from mouse splenocytes, bone marrow, or peripheral blood. The kit is designed for processing up to 1 x 10^9 cells and targets hematopoietic mouse cells for removal using biotinylated antibodies recognizing specific cell surface markers. Unwanted cells are labeled with biotinylated antibodies and streptavidin-coated magnetic particles, then separated without columns using an EasySep™ magnet, with desired cells simply poured off into a new tube.

The workflow is fast, easy-to-use, and column-free, yielding isolated cells that are immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction. Components include an Isolation Cocktail (a combination of monoclonal antibodies in PBS and 0.1% BSA) and EasySep™ Streptavidin RapidSpheres™ (a suspension of magnetic particles in PBS), both stored at 2 - 8°C, along with mycoplasma-free Normal Rat Serum stored at -20°C. When using single-cell suspensions from other tissue types, the kit may require optimization.

In a typical example provided by the manufacturer, human peripheral blood mononuclear cells (PBMCs) seeded into mouse bone marrow at an average starting frequency of 1.85% yielded a human PBMC content of 80 - 98% in the isolated fraction, with typically 3 - 4.5 log depletion of targeted mouse CD45+ cells. In a separate data example with a starting frequency of 0.85%, the human PBMC content of the isolated fraction was 93.7%.
