EasySep Mouse CD49b Pos Sel Kit

STEMCELL Technologies offers an immunomagnetic positive selection kit for isolating highly purified CD49b+ cells from mouse splenocytes or other single-cell suspensions. The column-free workflow uses antibodies and magnetic particles together with an EasySep™ magnet, allowing unwanted cells to be poured off while desired cells remain in the tube. Isolated cells are immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction.

Designed for processing 2 x 10^9 cells, the kit achieves up to 90% purity, with the CD49b+ content of the isolated fraction typically ranging from 74 to 90%. In a representative example using mouse splenocytes, the purities of the start and final isolated fractions were 6.2% and 90.2%, respectively. Components include a PE Labeling Reagent (a combination of monoclonal antibodies in PBS, 0.1% BSA, and less than 0.1% sodium azide, including an Fc receptor blocking antibody), a PE Selection Cocktail (monoclonal antibodies in PBS and 0.1% BSA), and EasySep™ Dextran RapidSpheres™ (magnetic particles in water), all stored at 2 - 8°C.

Protocols are available for both manual use with an EasySep™ magnet and fully automated use on the RoboSep™ system. The recommended medium is EasySep™ Buffer, RoboSep™ Buffer, or PBS containing 2% FBS and 1 mM EDTA, free of Ca++ and Mg++. For spleen samples, cells are prepared in PBS or Hanks' Balanced Salt Solution with 2% FBS, passed through a 70 μm mesh strainer, centrifuged at 300 x g for 10 minutes, and resuspended at 1 x 10^8 nucleated cells/mL.
