Mouse CD4+ T cells can be enriched directly from single-cell suspensions using the immunomagnetic positive-selection chemistry developed by STEMCELL Technologies for the EasySep platform. The selection cocktail targets the CD4 surface glycoprotein through a tetrameric antibody complex that links target lymphocytes to dextran-coated magnetic particles, allowing untouched CD4+ T cells to be recovered in a single column-free separation step performed inside a compatible EasySep magnet. Common downstream uses include in vitro T cell activation and differentiation assays, adoptive transfer into recipient mice, Treg and Th17 subset characterization, flow cytometry analysis, and gene expression profiling of helper T cell populations.

A standard small-scale separation handles up to 1 × 10^8 cells in roughly 0.5 to 2 mL of suspension, with a 50 uL sample aliquot supported on the lowest-volume protocol for limiting-dilution work. Recovered cells remain functionally viable, with reported purities generally exceeding 90 percent and recoveries typically between 60 and 80 percent depending on tissue source such as spleen, lymph node, bone marrow, or peripheral blood mononuclear cells. Reagents are supplied in single-use vials and should be kept refrigerated between uses, protected from light until first opening, and equilibrated before each run.

Researchers handling rare samples can scale the protocol downward, while bulk isolations from whole-spleen preparations scale upward in larger reaction tubes using the same chemistry, making the kit adaptable across immunology, autoimmune disease models, infectious disease research, and vaccine development workflows where defined CD4+ starting populations are required.
