EasySep Mouse CD138 Pos Selection Kit

This immunomagnetic positive selection kit from STEMCELL Technologies isolates highly purified CD138+ cells from mouse splenocytes, lymph nodes, or bone marrow. The kit is designed to deliver up to 97% purity when used with cells from immunized mice, and is particularly useful for the enrichment of plasma cells. The workflow is column-free and is compatible with hybridoma generation protocols, including electrofusion.

The kit targets CD138+ cells via an antibody recognizing the CD138 surface marker. Desired cells are labeled with antibodies and magnetic particles and separated using an EasySep™ magnet, with unwanted cells simply poured off while the desired cells remain in the tube. Isolated cells are immediately available for downstream applications such as flow cytometry, cell culture, and hybridoma generation. The kit can process up to 2 x 10^9 cells and supports both manual and fully automated RoboSep™ protocols.

Typical isolation data demonstrate consistent performance across sample types. Starting with immunized BALB/c mouse splenocytes, the CD138+ cell content of the isolated fraction is typically 81.5 ± 4.9% (mean ± SD), and the plasma cell (CD138+CD267 (TACI)+) content is typically 68.5 ± 11.3%. Starting with naïve C57BL/6 mouse splenocytes, the CD138+ cell content is typically 78.3 ± 5.7%, and the plasma cell content is typically 50.8 ± 10.0%. In hybridoma generation experiments, isolated CD138+ cells yielded an antigen-specific hit rate of 28.8 ± 11.0% and an antibody-secreting hybridoma rate of 99.3 ± 0.6%, compared with 4.1 ± 1.6% and 33.0 ± 8.7% respectively for total splenocytes.
