From STEMCELL Technologies, this immunomagnetic positive selection system isolates highly purified CD11c+ cells from mouse splenocytes or cultured bone marrow cells. The column-free protocol uses antibodies recognizing the CD11c surface marker together with magnetic particles; labeled cells are separated using an EasySep™ magnet while unwanted cells are simply poured off. Isolated cells are not fluorochrome-labeled and are immediately available for downstream applications such as flow cytometry, culture, and cell-based experiments.

Purities of up to 95% are achievable. Starting with mouse splenocytes, the CD11c+ cell content of the enriched fraction is typically 86.8 ± 9.7% (gated on viable singlet cells, mean ± SD) using the purple EasySep™ Magnet, with example start and isolated fraction purities of 5.7% and 92.3%, respectively. Sample preparation uses a cell concentration of 1 x 10^8 nucleated cells/mL. Spleen Dissociation Medium is included for tissue processing. The kit provides a Selection Cocktail of monoclonal antibodies in PBS and 0.1% BSA, EasySep™ Dextran RapidSpheres™ magnetic particles, and Mouse FcR Blocker. Select EasySep™ Mouse kits have transitioned from Normal Rat Serum to a Mouse FcR Blocker, with all kit components now shipped in a single package.
