EasySep Human pDC Enrichment Kit

An immunomagnetic, column-free cell separation kit from STEMCELL Technologies, designed for processing 2 x 10^9 cells to isolate untouched and highly purified human plasmacytoid dendritic cells (pDCs) from fresh peripheral blood mononuclear cells (PBMCs) by negative selection. The kit targets non-pDCs, including myeloid dendritic cells (mDCs), for removal using antibodies that recognize specific cell surface markers. Unwanted cells are labeled with antibodies and magnetic particles and separated without columns using an EasySep™ magnet, allowing the desired cells to be poured off into a new tube.

According to the manufacturer, the kit isolates pDCs 33% faster than column-based methods, achieves purities up to 97% with high recovery, and delivers untouched, viable cells immediately available for downstream applications such as flow cytometry, culture, DNA/RNA extraction, viral antigen uptake and processing studies, T cell activation and cross priming experiments, and innate and adaptive immunity research. The product information sheet indicates that, starting with PBMCs containing 0.2 - 0.9% pDCs, the pDC content of the enriched fraction typically ranges from 87 - 97%. pDCs are described as Lineage (CD3, CD14, CD16, CD19, CD20, CD34, CD56)-negative, HLA-DR positive, and CD304 (BDCA-4)-positive.

The kit is supplied with an Enrichment Cocktail, EasySep™ D Magnetic Particles, and an Anti-Human CD32 (Fc gamma RII) Blocker, all stored at 2 - 8°C. The product is intended for research use only.
