EasySep Human NK Cell Iso Kit

From STEMCELL Technologies, this kit uses column-free immunomagnetic negative selection to isolate highly purified, untouched human NK cells from fresh or previously frozen peripheral blood mononuclear cells (PBMCs) or washed leukapheresis samples in as little as 8 minutes.

The NK cell (CD3−CD56+) content of the isolated fraction is typically 85.0 ± 8.0% (mean ± SD). In a representative experiment, start and isolated fractions measured 5.9% and 86.7%, respectively. Isolated CD56+CD3− NK cells can be expanded over 14 days in feeder- and serum-free culture conditions using the ImmunoCult™ NK Cell Expansion Kit, with an average fold expansion of 89 ± 17. On Day 14, the average frequencies of viable CD56+CD3− and CD56+CD16+ NK cells were 87 ± 1% and 75 ± 2%, respectively. The expanded NK cells remain functional: in a 4-hour co-culture with K562 target cells at a 1:1 ratio, an average of 48 ± 2.4% of K562 cells were killed.

The workflow supports downstream research applications, including immunomagnetic negative selection of untouched human NK cells directly from whole blood, and is compatible with the company's range of cell separation buffers and purity-assessment antibodies.
