EasySep Human Myeloid Pos Slctn Kit II

From STEMCELL Technologies, this immunomagnetic positive selection kit isolates highly purified human myeloid cells (CD33+ and CD66b+) from nucleated cell suspensions prepared from human whole blood or bone marrow. The kit targets myeloid cells with antibodies recognizing the CD33 and CD66b surface markers; desired cells are labeled with antibodies and magnetic particles, then separated without columns using an EasySep™ magnet. Unwanted cells are poured off while desired cells remain in the tube.

Designed for processing 1 x 10⁹ cells, the kit requires no columns and can deliver up to 99% purity. Isolated cells are immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction. Sample preparation involves lysing whole blood or bone marrow to create a nucleated cell suspension, then resuspending cells at 1 x 10⁸ cells/mL in recommended medium (EasySep™ Buffer, RoboSep™ Buffer, or PBS containing 2% fetal bovine serum and 1 mM EDTA).

Starting with peripheral blood mononuclear cells, the myeloid cell content (CD14+CD66b+) of the isolated fraction is typically 96.5 ± 1.3% (mean ± SD using the purple EasySep™ Magnet); in the cited example, start and final isolated fraction purities were 59.2% and 94.6%, respectively. The included anti-CD33 antibody clone may block some anti-CD33 clones used to assess purity by flow cytometry; recommended fluorochrome-conjugated detection antibodies include anti-CD14 clones M5E2 or MoP9 and anti-CD66b clone G10F5.
