EasySep Human EpCAM Pos Slctn Kit II

STEMCELL Technologies offers a column-free immunomagnetic kit for isolating highly purified human EpCAM+ cells, such as mammary epithelial cells, from fresh or previously frozen cultured human mammary epithelial cells or other dissociated tissue samples. The positive selection approach uses an antibody recognizing the EpCAM surface marker to label desired cells with magnetic particles, which are then separated without columns using an EasySep™ magnet. Unwanted cells are simply poured off while desired cells remain in the tube, and isolated cells are immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction. The kit supports research workflows including the culture and evaluation of human mammary epithelial cells in CFU assays, the culture of human mammospheres and tumorspheres, and the general culture of human mammary epithelial cells.

The kit is designed to process up to 1 x 10^9 cells and is supplied as a combination of monoclonal antibodies in PBS (including an Fc receptor blocking antibody) together with EasySep™ Dextran RapidSpheres™ magnetic particles. Both components should be stored at 2 - 8°C, must not be frozen, and are stable until the expiry date on the label; components may be shipped at room temperature (15 - 25°C) but should be stored as indicated. According to the manufacturer's data, starting with peripheral blood mononuclear cells seeded with MCF-7 cells at a starting frequency of 10.8 - 50.0%, the EpCAM+ cell content (epithelial cell+CD45-) of the isolated fraction is typically 96.2 ± 3% (mean ± SD using the purple EasySep™ Magnet), and the documentation indicates that purities of up to 99% are achievable.

For purity assessment by flow cytometry, the manufacturer recommends the Anti-Human Epithelial Cell Antibody, Clone 5E11.3.1 (whose 5E11 epitope has an identical distribution to EpCAM) together with an Anti-Human CD45 Antibody, Clone HI30. The recommended medium is Hanks' Balanced Salt Solution (HBSS) modified with 10 mM HEPES, without phenol red, containing 2% fetal bovine serum. The kit can be used manually with EasySep™ magnets or run fully automated on the RoboSep™ instrument.
