EasySep Human CD4 Pos Sel Kit II

STEMCELL Technologies offers a column-free immunomagnetic positive selection kit designed to isolate highly purified human CD4+ cells from fresh or previously frozen peripheral blood mononuclear cells (PBMCs) or washed leukapheresis samples in as little as 15 minutes. The process uses monoclonal antibodies that recognize the CD4 surface marker along with magnetic particles, and an EasySep™ magnet retains desired cells while unwanted cells are poured off.

Components supplied are an EasySep™ Human CD4 Positive Selection Cocktail II (a combination of monoclonal antibodies in PBS with 10% HPCD) and EasySep™ Dextran RapidSpheres™ (a suspension of magnetic particles in water), both stored at 2 - 8°C and not to be frozen. Isolated cells are immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction.

Reported CD4+ T cell purity in the isolated fraction varies by starting material: starting with frozen mononuclear cells, it typically ranges from 92% - 97%; starting with fresh human PBMCs, it is typically 96.3 ± 3.0% (mean ± SD using the purple EasySep™ magnet), with another dataset reporting 90 ± 6%. The product information sheet notes that the selection cocktail uses an anti-CD4 antibody clone that may block other anti-CD4 antibody clones used to assess purity by flow cytometry, and recommends the Anti-Human CD4 Antibody, Clone OKT4, or alternative markers such as CD3 and CD8a. The kit is compatible with multiple EasySep™ magnets and the automated RoboSep™ platform, is intended for research use only, and is manufactured under a quality management system certified to ISO 13485.
