EasySep Hu WB CD34 Pos Slctn Kit II

A column-free immunomagnetic kit from STEMCELL Technologies enables positive selection of human CD34+ cells directly from fresh whole blood or buffy coat. Target cells are labeled using a complex that binds CD34 together with dextran-coated magnetic particles, and the antibody cocktail includes antibodies against human Fc receptors to limit nonspecific binding. Unwanted non-CD34+ cells are poured off while CD34+ cells remain in the tube, using an EasySep™ magnet rather than a column. CD34 is expressed on hematopoietic stem and progenitor cells.

When starting with whole blood, the CD34+ content of the isolated fraction is typically 90.4 ± 7.0% (gated on CD45+ cells; mean ± SD using RoboSep™-S); in the cited example, start and final isolated fractions were 0.06% and 95.31% respectively, with RBCs removed by lysis prior to flow cytometry. The kit forms part of a next-generation EasySep™ line intended to match or exceed the performance of earlier kits while shortening protocol times, and is compatible with the RoboSep™-S automated cell separation instrument for fully automated cell separation. A mouse monoclonal IgG1 antibody against human CD34 is offered for purity assessment of the isolated fraction.
