EasySep Hu Th17 Cell Enr Kit II

Designed for positive selection of human Th17 cells, this column-free immunomagnetic kit from STEMCELL Technologies isolates magnetic particle-free, highly purified CD4+CXCR3-CCR6+ Th17 cells from fresh peripheral blood mononuclear cells (PBMCs) or leukapheresis samples. The fast and easy-to-use, two-step procedure first isolates CCR6+ cells using antibody complexes and Releasable RapidSpheres™, then removes bound magnetic particles and depletes unwanted non-CD4+ T cells, CD45RA+ cells, and CXCR3+ cells using a CD4+CXCR3- T Cell Pre-Enrichment Cocktail and Dextran RapidSpheres™. The resulting isolated fraction is enriched for IL-17-producing Th17 cells, ready for downstream applications.

The kit is rated for up to 98% purity and is intended for processing up to 2 x 10^9 cells. In typical enrichments, Th17 cell content in the isolated fraction ranges from 96 - 98%. After overnight stimulation with PMA-Ionomycin, 6 - 19% of isolated cells are IL-17+ by intracellular flow cytometry, and IFN-γ-producing cells are typically less than 5% of the enriched fraction. Included components are a Human CCR6 Positive Selection Cocktail II, a Human CD4+CXCR3- T Cell Pre-Enrichment Cocktail, Releasable RapidSpheres™, Dextran RapidSpheres™, and Release Buffer, all stored at 2 - 8°C.
