EasySep Hu TCR Alpha/Beta Depl Kit

This immunomagnetic negative selection kit from STEMCELL Technologies is designed to deplete human T cell receptor alpha/beta+ (TCRαß+) cells from leukapheresis samples or expanded TCRαß knockout cell cultures. The kit targets TCRαß+ cells with an antibody recognizing the TCRαß surface marker; unwanted cells are labeled with antibody and magnetic particles and separated without columns using an EasySep™ magnet. Desired cells are simply poured off into a new tube, and isolated cells are immediately available for downstream applications such as flow cytometry, culture, or cryopreservation.

Key features include fast and easy-to-use operation, no columns required, and untouched isolated cells. The kit is intended for processing 1 x 10^9 cells, and the depletion protocol can be completed in 13 minutes. Components comprise the EasySep™ Human TCR Alpha/Beta Depletion Cocktail, a combination of monoclonal antibodies in PBS, and EasySep™ Dextran RapidSpheres™, a suspension of magnetic particles in water, both stored at 2 - 8°C and not to be frozen. According to the manufacturer, TCRαß-knockout efficiency typically ranges from 60 - 90% when disruption is carried out with the ArciTect™ CRISPR/Cas9 system. In a representative dataset, the frequencies of CD3+TCRαß+ cells in the starting and depleted fractions were 19.1% and 0.1%, respectively.
