EasySep Hu pDC Iso Kit

From STEMCELL Technologies, this kit enables isolation of highly purified, untouched human plasmacytoid dendritic cells (pDCs) from fresh peripheral blood mononuclear cells (PBMCs) or leukapheresis samples using immunomagnetic negative selection. The column-free workflow labels unwanted cells — including myeloid dendritic cells (mDCs) — with antibodies and magnetic particles, which are then separated using an EasySep™ magnet; the desired pDCs are simply poured or pipetted off into a new tube and remain immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction.

Up to 97% purity is reported for isolated cells, and the kit is designed for processing 2 x 10^9 cells. Starting with leukapheresis samples, the pDC content (Lin-HLA-DR+CD123+) of the isolated fraction is typically 90 ± 5.3% (mean ± SD) using the silver "Big Easy" EasySep™ Magnet, with a representative example showing start and final isolated fractions of 0.2% and 89.8%. Isolated pDCs are described as Lineage-negative (CD3, CD14, CD16, CD19, CD20, CD34, CD56), HLA-DR positive, and CD123 (IL-3Rα) positive.

Components include a Plasmacytoid DC Isolation Cocktail (Components A and B), Dextran RapidSpheres™ magnetic particles, and an Anti-Human CD32 (Fc gamma RII) Blocker, all to be stored at 2 - 8°C without freezing. The kit is compatible with multiple EasySep™ magnet platforms, including "The Big Easy," EasyEights™, Easy 50, and the fully automated RoboSep™, and is intended for research use only.
