EasySep Hu Pan-Granulocyte Isolation Kit

From STEMCELL Technologies, this immunomagnetic negative selection kit isolates highly purified pan-granulocytes — including neutrophils, eosinophils, and basophils — from fresh human peripheral blood leukocytes. The column-free workflow uses the EasySep platform: unwanted non-granulocytes are labeled with monoclonal antibodies and magnetic particles, then separated with an EasySep magnet so the desired cells can simply be poured off into a new tube.

Starting with whole peripheral blood, the total granulocyte content of the isolated fraction typically ranges from 97–99%, with reported purity of up to 99%. Isolated cells are untouched and viable, ready immediately for downstream applications such as flow cytometry, cell culture, or DNA/RNA extraction. Two sample preparation workflows are supported: an ammonium chloride RBC lysis method (preferred for slightly higher recovery) and a HetaSep RBC sedimentation method (preferred for faster, lysis-free sample processing). The kit is rated for processing 1 x 10^9 cells and is compatible with the EasySep, "The Big Easy," and Easy 50 magnets, as well as the fully automated RoboSep system. Components are stored at 2–8°C and must not be frozen.

Purity can be assessed by flow cytometry using anti-human CD16 (Clone 3G8) and anti-human CD66b (Clone G10F5), with optional anti-human CD123 (Clone 6H6), anti-human IgE, and anti-human CD203c antibodies for basophil characterization.
