Description

The kit from STEMCELL Technologies enables isolation of highly purified human extracellular vesicles (EVs) from plasma, serum, cell culture conditioned medium, and urine samples using immunomagnetic positive selection in less than 30 minutes. It is designed for processing 20 mL of biofluid. The kit targets EVs for positive selection with antibodies recognizing the tetraspanin markers CD9, CD63, and CD81. Desired EVs are labeled with antibodies and magnetic particles and separated without columns using an EasySep™ magnet, with unwanted biofluid components simply poured off while desired EVs remain in the tube. Following positive selection, particles should not be released from EVs, and the final isolated fraction contains highly purified EVs immediately available for downstream applications such as DNA/RNA extraction, Western blot, or mass spectrometry.

The selection cocktail is a combination of monoclonal antibodies in PBS, and the magnetic particles are a suspension in water. Both components should be stored at 2 - 8 °C, should not be frozen, and remain stable until the expiry date on the label. Western blot analyses show positive isolation of EVs with CD9, CD63, and CD81 tetraspanin markers from human plasma and mesenchymal stromal cell-conditioned medium. Transmission electron microscopy analysis following EasySep™ positive selection shows intact, spherical plasma-derived EVs. Proteomic analysis and RT-qPCR demonstrated compatibility of isolated EVs with mass spectrometry analysis and RNA analysis, respectively. The kit achieves equal or higher recovery of EVs from mesenchymal stromal cell-conditioned MesenCult™-ACF Plus Medium and plasma compared to other commercially available immunocapture-based EV isolation kits, and produces higher EV yield and purity from urine samples compared to differential ultracentrifugation.
