EasySep Hu Pan-CD25 Pos & Depl Kit

STEMCELL Technologies offers a kit for isolating or depleting CD25+ cells from fresh human peripheral blood mononuclear cells (PBMCs) or lysed leukapheresis samples by immunomagnetic selection. The workflow is column-free, using an EasySep™ magnet: cells are labeled with antibodies and magnetic particles, and unlabeled cells are simply poured off while labeled cells remain in the tube. The protocol is fast and easy-to-use, and the kit is rated for processing 1 x 10^9 cells.

Performance is reported as up to 1.3 log depletion of CD25+ cells and up to 98% purity. With fresh PBMCs, the depleted fraction typically contains 2 - 5% CD25+ cells, and the isolated fraction from the positive selection protocol typically ranges from 81 - 98% purity. Isolated cells are immediately available for downstream applications including flow cytometry, in vitro assays, and cell culture.

Components include a combination of monoclonal antibodies in PBS containing an Fc receptor-blocking antibody, plus a suspension of magnetic particles in water, both stored at 2 - 8°C. The protocol is designed to work with the EasySep™ magnet and "The Big Easy" magnet. The included antibody cocktail recognizes epitope B of the CD25 antigen, so for purity assessment by flow cytometry, fluorochrome-conjugated antibody clones recognizing epitope A are recommended.
