Built around the column-free EasySep platform, this immunomagnetic positive selection kit from STEMCELL Technologies isolates human myeloid-lineage cells, including CD33-positive monocytes and conventional dendritic cells, from whole blood, peripheral blood mononuclear cells, apheresis products, or bone marrow in a single sterile tube.

The workflow relies on antibody-coated magnetic nanoparticles that label the surface markers on target cells. After a brief incubation, the tube is placed in the EasySep magnet, and labeled myeloid cells are retained against the wall while unwanted cells are decanted. Because separation happens entirely inside a tube rather than on a column, the protocol preserves cell viability, surface marker expression, and function for downstream applications including flow cytometry, dendritic cell differentiation, cell culture, and functional immunology studies.

Each labeling reaction processes up to 25 mL of whole blood, or up to 4 x 10^9 cells from a leukocyte-rich suspension, and reproducibly delivers purity greater than 90% alongside yields exceeding 50% of the target cells in the starting sample. The kit format pairs a human-specific antibody cocktail with magnetic nanoparticle reagent, both supplied in single-use vials.
