EasySep Hu Monocyte Iso Kit, 10 mL

This immunomagnetic negative selection kit from STEMCELL Technologies isolates untouched and highly purified CD14+CD16- monocytes from fresh leukapheresis samples. The kit targets non-monocytes and CD16+ monocytes for removal using antibodies that recognize specific cell surface markers; unwanted cells are labeled with antibodies and magnetic particles and separated without columns using an EasySep™ magnet. Desired cells are simply pipetted off into a new flask and are immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction. The workflow is fast, easy to use, and column-free, yielding untouched, viable cells with up to 90% purity and high recovery. The kit is designed for processing 1 x 10^10 cells using the Easy 250 EasySep™ Magnet.

Two components are supplied: the EasySep™ Human Monocyte Isolation Cocktail, a combination of monoclonal antibodies in PBS that includes an Fc receptor blocking antibody, and EasySep™ D Magnetic Particles, a suspension of magnetic particles in TBS. Both components should be stored at 2 - 8°C, must not be frozen, and are stable until the expiry date on the label; they may be shipped at room temperature (15 - 25°C). Recommended medium is EasySep™ Buffer, RoboSep™ Buffer, or PBS containing 2% FBS and 1 mM EDTA, and should be free of Ca++ and Mg++.

Starting with washed or lysed leukapheresis samples, the monocyte cell content (CD14+CD16-) of the isolated fraction is typically 83.6 ± 6.0% (gated on CD45, mean ± SD for the Easy 250 EasySep™ Magnet). For purity assessment of CD14+CD45+ monocytes by flow cytometry, anti-human CD14 antibody (Clone M5E2) and anti-human CD45 antibody (Clone HI30) are recommended.
