EasySep Hu Memory B Cell Iso Kit

STEMCELL Technologies' EasySep Human Memory B Cell Isolation Kit is designed to isolate highly purified CD27+ memory B cells from fresh or previously frozen human peripheral blood mononuclear cells (PBMCs) or leukapheresis samples. Isolation is achieved in as little as 40 minutes using column-free immunomagnetic positive selection, with the kit rated for processing 2 x 10^9 cells. The workflow first enriches CD27+ cells using antibody complexes together with EasySep Releasable RapidSpheres, after which the bound magnetic particles are released from the selected cells. Unwanted non-B cells are then depleted using antibody complexes and EasySep Dextran RapidSpheres, yielding a final fraction of highly purified memory B cells ready for downstream applications.

An optional protocol allows untouched naïve B cells to be isolated in parallel from the same sample for functional studies. Using the purple EasySep magnet, the memory B cell content (CD19+CD27+) of the isolated fraction is typically 97 ± 2% (mean ± SD), while the naïve B cell content (CD19+CD27-) obtained with the optional protocol is typically 93 ± 5% (mean ± SD).

The procedure is performed manually using a compatible EasySep magnet. Recommended sample preparation includes resuspending cells at 1 x 10^8 cells/mL in EasySep Buffer or PBS containing 2% fetal bovine serum and 1 mM EDTA, in a medium free of Ca++ and Mg++.
