EasySep Hu Gamma Delta T Cell Iso Kit

From STEMCELL Technologies, this kit uses immunomagnetic negative selection to isolate untouched and highly purified human gamma/delta T cells from fresh or previously frozen peripheral blood mononuclear cells (PBMCs) or from lysed leukapheresis samples. Non-gamma/delta T cells are labelled with antibodies and magnetic particles, then separated without columns using an EasySep™ magnet, leaving the desired cells to be poured off into a new tube.

The protocol is fast, easy to use, and column-free, with isolated cells immediately ready for downstream applications such as flow cytometry, culture, or DNA/RNA extraction. Starting from fresh PBMCs, the gamma/delta TCR+CD3+ cell content of the isolated fraction typically ranges from 90 - 97%, with up to 97% purity reported. For purity assessment by flow cytometry, recommended antibodies include an Anti-Human CD3 Antibody, Clone UCHT1, together with an anti-human TCR gamma/delta antibody (e.g., clone IMMU510 or 11F2).

Compatible separation platforms include the EasySep™ Magnet, "The Big Easy" EasySep™ Magnet, Easy 50 EasySep™ Magnet, and RoboSep™-S. The Isolation Cocktail is a combination of monoclonal antibodies in PBS, and the magnetic particles are supplied as a suspension in TRIS-buffered saline. Both components are stored at 2 - 8 °C and should not be frozen. The recommended medium is EasySep™ Buffer, RoboSep™ Buffer, or PBS containing 2% fetal bovine serum and 1 mM EDTA, free of Ca²⁺ and Mg²⁺.
