EasySep Hu Cen/Eff Mem CD8T Cell Iso Kit

This immunomagnetic isolation kit from STEMCELL Technologies enables researchers to obtain enriched human central and effector memory CD8+ T cells. Using particle release technology, the kit isolates central memory CD8+ T cells (CD3+CD8+CD45RO+CD62L+) and effector memory CD8+ T cells (CD3+CD8+CD45RO+CD62L-) from human PBMCs or washed leukapheresis samples in as little as 43 minutes, in a column-free workflow.

According to the manufacturer, when starting with fresh PBMCs, the central memory CD8+ T cell content (CD3+CD8+CD45RO+CD62L+) of the isolated fraction is typically 86.8 ± 8.4% (mean ± SD using the purple EasySep™ magnet). The effector memory content (CD3+CD8+CD45RO+CD62L-) of the isolated fraction is typically 88.7 ± 6.5% (mean ± SD). RBCs were removed from the start sample by lysis prior to flow cytometry.

The workflow uses an antibody cocktail together with EasySep™ Releasable RapidSpheres™ for positive selection of CD62L+ cells, after which the magnetic particles are released. Unwanted CD45RA+ cells and non-CD8+ T cells are then labelled and depleted using EasySep™ Dextran RapidSpheres™, and effector memory CD8+ T cells can be recovered from the CD62L– fraction.
