EasySep Hu CD8 Pos Sel Kit II, 10 mL

From STEMCELL Technologies, the EasySep™ Human CD8 Positive Selection Kit II enables isolation of highly purified human CD8+ T cells from PBMCs or washed leukapheresis samples by immunomagnetic positive selection, achieving up to 99% purity with high recovery. The kit uses antibodies recognising the CD8 surface marker together with magnetic particles; desired cells are labelled and separated without columns using an EasySep™ magnet, with unwanted cells simply pipetted off while the CD8+ fraction remains in the flask. Isolated cells are immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction.

The kit is designed for processing 1 × 10^10 cells when used with the Easy 250 EasySep™ Magnet. It contains the EasySep™ Human CD8 Positive Selection Cocktail II — a combination of monoclonal antibodies in PBS with 10% HPCD and 0.09% rHA, including an Fc receptor blocking antibody — and EasySep™ Dextran RapidSpheres™, a suspension of magnetic particles in water. Both components are stored at 2 – 8 °C and should not be frozen.

Starting from washed or lysed leukapheresis samples, the CD8+ T cell content (CD3+CD8+) of the isolated fraction is typically 93.9 ± 4.9% (gated on viable cells, mean ± SD for the Easy 250 EasySep™ Magnet). Using "The Big Easy" EasySep™ Magnet with a single-cell suspension of human PBMCs, the CD8+ cell content of the isolated fraction is typically 96.5 ± 2.4% (mean ± SD). CD8 is expressed on cytotoxic T cells and weakly on a subset of NK cells. Purity can be assessed by flow cytometry using a fluorochrome-conjugated anti-human CD8a antibody such as Clone RPA-T8.
