EasySep Hu CD45 Depletion Kit II

The kit from STEMCELL Technologies depletes human CD45+ cells from fresh or previously frozen peripheral blood mononuclear cells (PBMCs) through a column-free immunomagnetic separation workflow. It uses a monoclonal antibody combination recognizing the CD45 cell surface marker, along with magnetic particles, to label and remove unwanted cells without columns. Desired cells are poured off into a new tube and remain untouched, immediately ready for downstream applications such as flow cytometry, culture, or DNA/RNA extraction.

The kit is rated for processing 2 × 10^9 cells and achieves up to a 4.0 log depletion of CD45+ cells; in a representative profile, CAMA cells seeded into PBMCs at a starting frequency of 1.1% (98.9% CD45+) were recovered at 98% in the depleted fraction. The included antibody reagent includes a mouse monoclonal IgG1 directed against human and chimpanzee CD45. Protocols are provided for both manual EasySep™ magnets and the fully automated RoboSep™ system. Components are the EasySep™ Human CD45 Depletion Cocktail II (a combination of monoclonal antibodies in PBS) and EasySep™ Dextran RapidSpheres™ (a suspension of magnetic particles in water).
