EasySep Hu CD4+ T Cell Iso Kit, 10mL

From STEMCELL Technologies, this immunomagnetic negative selection kit is designed to isolate untouched and highly purified CD4+ T cells from fresh leukapheresis samples. The process is column-free, using an EasySep™ magnet to remove unwanted non-CD4+ T cells that have been labelled with antibodies and magnetic particles; the desired CD4+ T cells are simply pipetted off into a new flask. Isolated cells remain viable and are immediately ready for flow cytometry, culture, or DNA/RNA extraction.

The kit includes a Human CD4+ T Cell Isolation Cocktail, supplied as a combination of monoclonal antibodies in PBS, together with Dextran RapidSpheres™, a suspension of magnetic particles in water. Both components should be stored at 2 - 8°C and must not be frozen. For optimal performance, fresh lysed leukapheresis samples are recommended, with cells prepared at 5 x 10^7 cells/mL in the recommended medium.

Using the Easy 250 EasySep™ Magnet, the kit achieves up to 98% purity with high recovery. Starting from washed or lysed leukapheresis samples, the CD4+ T cell content (CD3+CD4+) of the isolated fraction is typically 96.5 ± 1.7%, gated on viable cells (mean ± SD). Starting from human peripheral blood mononuclear cells (PBMCs), the CD4+ T cell content is typically 94.8 ± 2.3% (mean ± SD), and isolation can be performed in as little as 8 minutes. Purity assessment by flow cytometry can be carried out using Anti-Human CD3 Antibody, Clone UCHT1, and Anti-Human CD4 Antibody, Clone OKT4.
