EasySep Hu CD4 Pos Sel Kit II, 5 mL

STEMCELL Technologies' EasySep™ kit uses immunomagnetic positive selection to isolate highly purified human CD4+ cells from leukapheresis samples. The kit targets CD4+ cells with antibodies recognizing the CD4 surface marker, labeling target cells with antibodies and magnetic particles that are separated without columns using an EasySep™ magnet. Unwanted cells are pipetted off while desired cells remain in the flask, and isolated cells are immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction.

The kit is designed for processing 1 x 10^10 cells using the Easy 250 EasySep™ Magnet and achieves up to 98% purity with high recovery. Starting with washed or lysed leukapheresis samples, the CD4+ T cell content (CD4+CD45+) of the isolated fraction is typically 96.4 ± 1.6% (gated on CD45, mean ± SD for the Easy 250 EasySep™ Magnet). When isolating from PBMCs or washed leukapheresis samples with the purple EasySep™ magnet, the CD4+ T cell content (CD3+CD4+) of the isolated fraction is typically 96.3 ± 3.0%, and isolation can be completed in as little as 15 minutes.

The Positive Selection Cocktail comprises monoclonal antibodies in PBS with 10% HPCD and 0.09% recombinant human albumin. Components should be stored at 2 - 8°C, not frozen, and remain stable until the expiry date on the label. For purity assessment by flow cytometry, Anti-Human CD4 Antibody, Clone OKT4 is recommended, since the cocktail's anti-CD4 clone may block other anti-CD4 antibody clones used for assessment.
