Description
From STEMCELL Technologies, this column-free, positive-selection immunomagnetic kit isolates human CD4+CD127lowCD25+ regulatory T cells (Tregs) from fresh or previously frozen peripheral blood mononuclear cells (PBMCs) or leukapheresis samples. CD25+ cells are first captured using EasySep™ Releasable RapidSpheres™; bound particles are then released, and unwanted non-Tregs are depleted, yielding a final fraction of highly purified CD4+CD127lowCD25+ cells that express high levels of FOXP3 and are immediately ready for downstream applications.
According to the manufacturer, processing up to 1 x 10^9 cells per isolation typically yields a fraction containing 85.0 ± 4.8% (mean ± SD) CD4+CD25+FOXP3+ Tregs, with the workflow completed in less than 1 hour. An optional parallel protocol allows CD4+CD25- responder T cells to be isolated from the same sample for functional studies. Following isolation, antibody complexes remain bound to the cell surface and may interact with Brilliant Violet™ antibody conjugates, polyethylene glycol-modified proteins, or other chemically related ligands.
Supplied components include a CD25 Positive Selection Cocktail, a CD4+ T Cell Enrichment Cocktail, a CD127high Depletion Cocktail, EasySep™ Releasable RapidSpheres™, EasySep™ Dextran RapidSpheres™ and EasySep™ Release Buffer. All components are stored at 2–8 °C and must not be frozen.
