EasySep Hu CD3 Pos Sel Kit II, 10 mL

From STEMCELL Technologies, this large-format kit is designed for column-free immunomagnetic positive selection to isolate highly purified CD3+ cells from leukapheresis samples. Cells are labeled with a cocktail of monoclonal antibodies (including an Fc receptor blocking antibody) and magnetic particles, then separated in a flask using an EasySep™ magnet. Unwanted cells are simply pipetted off while desired cells remain in the flask, with no columns required. The kit is intended for processing up to 1 x 10^10 cells using the Easy 250 EasySep™ Magnet, and isolated cells are immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction.

Two components are supplied: a Positive Selection Cocktail II — a combination of monoclonal antibodies in PBS with 10% HPCD and 0.09% rHA — and EasySep™ Dextran RapidSpheres™, a suspension of magnetic particles in water. Both should be stored at 2 - 8°C and not frozen; components may be shipped at room temperature (15 - 25°C). The recommended medium is EasySep™ Buffer, RoboSep™ Buffer, or PBS containing 2% FBS and 1 mM EDTA, and should be free of Ca++ and Mg++.

Manufacturer performance figures report up to 99% purity with high recovery. Typical CD3+ cell content of the isolated fraction is given as 95.9 ± 2.8% (gated on viable cells, mean ± SD) when starting from washed or lysed leukapheresis samples on the Easy 250 EasySep™ Magnet, and 99.2 ± 0.2% (mean ± SD) when starting from PBMCs on the purple EasySep™ Magnet.
