EasySep Hu CB CD34 Pos Sel Kit III

This immunomagnetic positive selection kit from STEMCELL Technologies is designed to isolate highly purified human CD34+ cells from fresh whole umbilical cord blood using a simple, two-step procedure that combines RosetteSep™ pre-enrichment with EasySep™ magnetic isolation.

The RosetteSep™ antibody cocktail binds to T cells, B cells, red blood cells (RBCs), and myeloid cells, forming dense immunorosettes that are separated by density gradient centrifugation. Because the cocktail does not label platelets, the kit is recommended for fresh cord blood samples less than 24 hours after delivery, samples containing very few platelets, or when platelet depletion is not desired. Pre-enrichment can be carried out in SepMate™ tubes, which require only 10 minutes of centrifugation with the brake on. The pre-enriched CD34+ cells are then labeled with an EasySep™ isolation cocktail and magnetic EasySep™ Dextran RapidSpheres™, and positively selected by placing the tube into an appropriate EasySep™ magnet or RoboSep™ instrument.

Typical performance data obtained with the purple EasySep™ Magnet show a starting CD34+ cell content of 0.5 ± 0.25% (mean ± SD) and an isolated fraction purity of 87 ± 12% (mean ± SD, n=10). Across 10 different fresh cord blood samples, average purity progressed from 0.5% at start to 11% after RosetteSep™ pre-enrichment and 87% after EasySep™ selection, with ranges of 0.1 - 0.9%, 4 - 21%, and 56 - 99%, respectively. Each kit contains reagents sufficient for processing up to 1000 mL of cord blood and is intended for research use only.
