EasySep HLA Chim WB CD4 Pos Slctn Kit

STEMCELL Technologies offers an immunomagnetic positive selection kit that isolates highly purified human CD4+ cells from fresh whole blood, buffy coat, or leukocyte reduction system (LRS) cones. Using a column-free EasySep™ protocol, the kit labels target cells with antibodies and magnetic particles so that unwanted cells can be poured off while CD4+ cells remain in the tube. Starting from human whole blood, the CD4+CD14- content of the isolated fraction is typically 97.0 ± 1.8% (gated on CD45; mean ± SD using "The Big Easy" EasySep™ Magnet), with example start and final isolated-fraction purities of 29.8% and 99.0%, respectively.

Designed to process up to 60 mL of buffy coat or whole blood, the kit combines an EasySep™ HLA Chimerism Whole Blood CD4 Positive Selection Cocktail with EasySep™ Dextran RapidSpheres™ magnetic particles and a 10X concentrated Red Blood Cell Lysis Buffer. It is compatible with manual workflows using "The Big Easy" or EasyEights™ magnets and with the fully automated RoboSep™ platform. Isolated cells are ready for downstream applications such as flow cytometry, culture, or DNA/RNA extraction for lineage-specific chimerism analysis.

For purity assessment by flow cytometry, STEMCELL Technologies recommends an anti-human CD4 antibody, clone L120, optionally paired with anti-human CD14 antibodies, clones M5E2 or MoP9, since the selection cocktail's anti-CD4 clone is known to block clones RFT4, SK3, SK4, RPA-T4, and 13B8.2.
