EasySep Direct Monocyte Iso Kit

Designed by STEMCELL Technologies, this kit enables researchers to isolate highly purified human CD14+ monocytes directly from whole blood using immunomagnetic negative selection, with no density gradient centrifugation, sedimentation, or lysis required. Unwanted cells, including non-monocytes and CD16+ monocytes, are labeled with antibodies and EasySep™ Direct RapidSpheres™ and separated using an EasySep™ magnet in a column-free workflow. Isolated cells are left untouched and are immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction.

Reported performance includes greater than 99.9% red blood cell depletion and up to 92% purity of isolated cells. Starting with human whole blood from normal healthy donors, the typical monocyte (CD14+) content of the non-lysed final isolated fraction is 82.2 ± 8.4% (gated on CD45) or 79.0 ± 10.1% (not gated on CD45).

The kit supports manual protocols with multiple magnets, including EasySep™, The Big Easy, EasyEights™, and Easy 50, as well as a fully automated workflow on RoboSep™. EDTA anticoagulation is important for performance, with K2EDTA or K3EDTA recommended, and recovery of desired cells decreases with samples older than 24 hours.
