EasySep Direct Hu Total Lymph Iso Kit

From STEMCELL Technologies, this immunomagnetic kit isolates highly purified human lymphocytes directly from whole blood, buffy coat, or spleen by negative selection. The single-step procedure depletes red blood cells and unwanted cells without density gradient centrifugation, sedimentation, or RBC lysis, leaving target cells untouched and immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction. Non-lymphocytes are labeled with antibodies and EasySep™ Direct RapidSpheres™, then separated using a compatible EasySep™ magnet.

Individual samples of 0.5 – 30 mL can be processed in as little as 20 minutes. The kit achieves > 99.9% RBC depletion without pre-processing, and the typical total lymphocyte purity of the isolated fraction is 96.7 ± 1.5% (gated on CD45). It is suitable for use in flow cytometric crossmatch (FCXM) assays and is compatible with the EasySep™, "The Big Easy", EasyEights™, and Easy 50 magnets, as well as the fully automated RoboSep™ platform. Up to 16 samples can be processed simultaneously when combined with the EasyEights™ magnet.

Components include an Isolation Cocktail of monoclonal antibodies in PBS and a RapidSpheres™ suspension of magnetic particles, both stored at 2 – 8°C. The recommended medium is D-PBS without calcium and magnesium. For optimal RBC depletion, peripheral blood should be collected using heparin or acid citrate dextrose (ACD) as anticoagulant and processed within 24 hours of collection.
