EasySep Direct Hu T Cell-RoboSep

The EasySep™ Direct Human T Cell Isolation Kit from STEMCELL Technologies isolates highly purified human T cells directly from human whole blood using immunomagnetic negative selection, with no density gradient centrifugation, sedimentation, or RBC lysis required. Non-T cells are labeled with antibodies and separated using EasySep™ Direct RapidSpheres™ and an EasySep™ magnet, leaving the desired T cells untouched and immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction.

Reported kit benefits include greater than 99.9% RBC depletion, up to 97% purity of isolated cells, and a fast, easy-to-use, column-free workflow. Typical T cell (CD3+) content of the non-lysed final isolated fraction is 95.3 ± 1.4% (gated on CD45) or 94.9 ± 1.5% (not gated on CD45). The kit comprises an Isolation Cocktail of monoclonal antibodies in PBS and RapidSpheres™ magnetic particles in PBS, with components stored at 2 - 8°C and not frozen.

For optimal RBC depletion, blood should be collected using heparin or acid citrate dextrose (ACD) as an anticoagulant; K2EDTA or K3EDTA are not recommended. Best recovery is achieved with unprocessed human whole blood; recovery decreases for samples older than 24 hours.
