EasySep Direct Hu NK Iso Kit

STEMCELL Technologies' EasySep™ Direct Human NK Cell Isolation Kit enables researchers to isolate highly purified human natural killer (NK) cells directly from whole blood by immunomagnetic negative selection, without the need for density gradient centrifugation, sedimentation, or lysis. The workflow achieves > 99.9% red blood cell (RBC) depletion and delivers up to 94% purity of isolated cells, with the procedure completed in as little as 20 minutes using a column-free format.

Unwanted non-NK cells are labeled with a cocktail of monoclonal antibodies recognizing human CD3, CD45, and CD56 (NCAM), then targeted using EasySep™ Direct RapidSpheres™ and an EasySep™ magnet. The isolated NK cells are untouched and immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction. The kit is designed for processing 100 mL of whole blood.

For optimal RBC depletion, blood should be collected using heparin or acid-citrate-dextrose (ACD) as an anticoagulant; K2EDTA and K3EDTA are not recommended. Unprocessed human whole blood gives the best recovery, and recovery of the desired cells decreases with samples older than 24 hours. In typical data from normal healthy donors, the NK cell (CD3-CD56+) content of the non-lysed final isolated fraction is 90.4 ± 4.0% (gated on CD45) or 88.8 ± 3.7% (not gated on CD45).
