Description
The kit from STEMCELL Technologies enables isolation of human naïve B cells directly from whole blood without density gradient centrifugation, sedimentation, or RBC lysis, using an immunomagnetic negative selection protocol. Unwanted non-naïve B cells are labeled with antibodies and magnetic particles (EasySep™ Direct RapidSpheres™), and separation is performed with an EasySep™ Magnet without the use of columns. The unlabeled naïve B cells are collected in a new tube and can be used directly in downstream applications such as flow cytometry, cell culture, or nucleic acid extraction.
Typical naïve B cell (CD19+CD27-) content of the non-lysed final isolated fraction from healthy donor whole blood is reported as 91.8 ± 3.6% when gated on CD45, or 82.4 ± 12.6% when not gated on CD45. In a representative example, the naïve B cell content was 5.1% in the lysed whole blood start sample and 91.5% (gated on CD45) or 90.0% (not gated on CD45) in the non-lysed final isolated fraction, with a starting frequency of 0.009% in the non-lysed whole blood.
Separation can be performed in as little as 20 minutes using compatible magnets such as the EasyEights, EasySep Magnet, "The Big Easy" EasySep Magnet, or Easy 50 EasySep Magnet. The kit utilizes a mouse monoclonal IgG1 antibody against human and chimpanzee CD19 and a mouse monoclonal IgG1 antibody against human and chimpanzee CD45.
