EasySep Direct Hu CD8 Iso Kit

Developed by STEMCELL Technologies, this immunomagnetic negative selection kit isolates highly purified human CD8+ T cells directly from whole blood without density gradient centrifugation, sedimentation, or red blood cell (RBC) lysis. The column-free workflow achieves > 99.9% RBC depletion and up to 87% purity of isolated cells, which remain untouched and ready for downstream applications such as flow cytometry, culture, or DNA/RNA extraction.

The protocol targets non-CD8+ T cells for removal using a cocktail of monoclonal antibodies and EasySep™ Direct RapidSpheres™, with separation performed using an EasySep™ magnet. Desired cells are simply collected into a new tube. The kit includes an EasySep™ Direct Human CD8+ T Cell Isolation Cocktail and EasySep™ Direct RapidSpheres™; both components are stored at 2–8°C and should not be frozen.

For optimal RBC depletion, blood should be collected with heparin or acid-citrate-dextrose (ACD) as anticoagulant; K2EDTA or K3EDTA is not recommended, and recovery decreases with samples older than 24 hours. Typical CD8+ T cell (CD3+CD8+) content of the non-lysed final isolated fraction is 82.4 ± 4.9% (gated on CD45) or 81.6 ± 4.9% (not gated on CD45), with an example run reaching 87.3% purity. The kit is intended for research use only.
