EasySep Direct Hu B-CLL Iso Kit

This STEMCELL Technologies kit enables the isolation of highly purified human B-CLL cells directly from whole blood using immunomagnetic negative selection, without the need for density gradient centrifugation, sedimentation, or lysis. The depletion cocktail is formulated specifically for chronic lymphocytic leukemia (CLL) samples and notably does not contain antibodies against CD43, a marker that can be expressed on malignant cells in CLL. The workflow delivers > 99.9% red blood cell depletion and produces untouched isolated cells that are immediately ready for downstream applications such as flow cytometry, culture, or DNA/RNA extraction. Unwanted non-B cells are labeled with antibodies and EasySep™ RapidSpheres™, then separated using an EasySep™ magnet; desired cells are simply collected into a new tube.

The kit includes two components: an Isolation Cocktail and EasySep™ Direct RapidSpheres™, both stored at 2 - 8°C and shipped at room temperature (15 - 25°C). The antibody panel includes mouse monoclonal antibodies recognising human CD19, CD20, CD45, and CD5. For optimal red blood cell depletion, the manufacturer recommends collecting blood using heparin or acid citrate dextrose (ACD) as an anticoagulant, and processing unprocessed human whole blood for best recovery; K2EDTA or K3EDTA are not recommended. The kit can process up to 100 mL of whole blood.

According to typical performance data, starting with human whole blood from normal healthy donors, the B cell (CD3-CD19+) content of the non-lysed final isolated fraction is 87.0 ± 7.6% when gated on CD45, or 84.1 ± 11.7% when not gated on CD45. The purity of isolated cells is typically higher when processing samples with an elevated starting frequency of B cells, such as CLL samples.
