EasySep Direct Hu B Cell Iso Kit

This immunomagnetic negative selection kit from STEMCELL Technologies isolates highly purified B cells directly from human whole blood without the need for density gradient centrifugation, sedimentation, or lysis. The procedure is fast, easy-to-use, and column-free, leaving the isolated cells untouched and immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction.

The kit targets non-B cells for removal using antibodies that recognize specific cell surface markers. Unwanted cells are labeled with antibodies and EasySep™ Direct RapidSpheres™, then separated with an EasySep™ magnet, after which the desired cells are simply collected into a new tube. The kit comprises an EasySep™ Direct Human B Cell Isolation Cocktail and EasySep™ Direct RapidSpheres™, both formulated in PBS and stored at 2 - 8°C, stable until the expiry date on the label. For optimal RBC depletion, blood should be collected using heparin or acid-citrate-dextrose (ACD) as an anticoagulant; K2EDTA or K3EDTA are not recommended. For best recovery, unprocessed human whole blood should be used, as recovery decreases with samples that are older than 24 hours.

Documented performance includes > 99.9% RBC depletion and up to 95% purity of isolated cells. In data from healthy donor whole blood, the typical B cell (CD3-CD19+) content of the non-lysed final isolated fraction is 95.3 ± 2.7% (gated on CD45) or 88.5 ± 11.5% (not gated on CD45).
